Study on Shoot Proliferation of Fine Strain (13-38) of Sweet Cherry

Abstract: The present paper reported a method for fast-proliferation of the fine strain (13-38) of sweet cherry by shoot tip culture and graftage of plantlets regenerated in vitro. Resting buds were cut from the strain to prepare bacteria-free explants. The shoot tip Clones were established by culture on the MS medium supplemented with 0.1-0.5mg/L 6-BA, 0.1mg/L IAA, and differentiation of buds in the MS medium with 1-5mg/L ZT, 0.1-0.25mg/L IAA. The 48-108-fold increase of the buds were achieved in culture after one year. The survival rate of these graftages is 20% to 70%. by the methods of bark graft and cut graft, on the four kinds of stocks The graft-affinity of the new stock strain 1090 is higher than the other stocks. There was a higher survival rate by treating with 200ppm NAA on the graft unions of scion and the stock. The survival plants grafted may be transplanted directly or cut off its buds as scions to get a second reproduction in spring of the next year.
