Establishment of Regeneration and Genetic Transformation System of Strawberry Leaf

Abstract: An ideal regeneration system from leaf of strawberry cultivar M14 was developed. 10.0 shoots per leaf disc were generated at the rate of 100% within 3 weeks. Histological observations indicated that the shoots regenerated mainly from calli. This regeneration system was used in genetic transformation mediated by Agrobacterium tumefaciens strain (EHA105) carrying the modified pMDG410 plasmid, which contains GUS and NPTⅡgene. Kanamycin_resistant shoots regenerated with the ratio of 14.5% were identified as transgenic shoots.
