Application of Degenerate Primers for Amplification of the Gene Encoding Glycerol-3-phosphate Acyltransferase by Polymerase Chain Reaction

Abstract: Using highly degenerate primers by RT-PCR, the conserved region in GPAT-gene was obtained from total RNA of squash (Cucurbita moschata). In the PCR reactions, the coneen- tration ratio between the degenerate primers was essential for influencing amplification efficiency. Utilizing deoxyinosine(dI) can decrease degenerate degree and enhance the specificity of PCR products and amplification efficiency.
