TISSUE CULTURE OF RHAPHIDOPHORA DECURSIVA SHOOT TIPS AND STEM NODES IN VITRO

Abstract: Shoot tips and annual or biennial stem nodes of Rhaphido phora decursiva (Roxb.) Schott have been cultured on Murashige and Skoog ( MS ) me-dium supplemanted with 6-BA, IAA, NAA (can be omitted), and 10 coconut milk, All the dormant buds in shoot tips and stem nodes are able to sprout. When the dormant buds grow out 4-5 cuphylla and the plants reach about 7cm long, they are excised from explants and transplanted to the fresh MS medium. As the aerial roots grow out, the complete and aseptic plantlets are obtained. The surface sterilization of biennial stem nodes, however, is relatively difficult.
