Abstract: With the isolated and purified cpDNA, a library of cloned genomic fragments can be generated and the isolation and the nucleotide sequence of a given gene can further be conducted. However, the most generally applicable procedures for preparing purified cpDNA are very diffi-cult in Vitis species through the experience of our research work. In this paper we have developed a procedure which has been proved to be succesful in isolating and purifying the cpDNA of Vitis species, bearing comparing some species of other family.
