PROTOPLAST ISOLATION AND ITS REGENERATION OF MORCHELLA CONICA

Abstract: Abstract Protoplasts of Morchella conica Pers were isolated from the mycelia cultured in liquid medium. The highest isolated quantity v/as 6.2×10^6 protoplasts/ 100mg·ml in mixed enzyme solution No. 2 and at 30°C tor 4.5 h in cubation. Isolated protoplasts regenerated to hyphae readily. After 18 hours of incubation hyphae grew out from the cells regenerated from the protoplasts. Following plating on solid medium regenerated isolates were obtained.
