CALLUS CULTURE FROM PANAX NOTOGINSENG

Abstract: Abstract The growth rate was 54.0 mg D.W./1/day and saponin content was 5.37% for original culture callus from Panax notoginseng, As we considered low growth rate of P. notoginseng callus and saponin content, the suitable KT concentration and 2, 4-D concentration should be 0.7 ppm and 2.5 ppm respectively when the different concentration of KT and 2, 4-D were added in to medium. Coconut milk (CM) and lactalbumin hydrolysatc (LH) had more efficent nutrient regulation in the callus culture. 20% CM and 0.7% LH were therefore more suitable both growth and saponin content respectively. Five fine clones had been selected from twenty-one clones of P. notoginseng callus, amony which No. 04 which growth rate was as high as 219.2 mg D.W./1/day and that saponin content 12.8 was finest. The growth rate of the callus was reached as high as 220 mg D.W./I/day through the studies on metabolism regulation in callus culture of P. notoginseng and this was four times as that of the original culture callus. The total saponin content in the callus was 13% which was 2.4 times as that the original culture callus and as three times of the original plant. The results confirmed that the totipotency of secondary metabolism in culture cell of P. notoginseng might be regulated.
