STUDIES ON TISSUE CULTURE AND TISSUE DIFFERENTIATI0N 0F SCAPE SEGNIENTS IN PULSATILLA CHINENSIS

Abstract: Different parts segments of scape in Pulsatilla chinensis were cultured. On MS medium supplcmented with 6-BA 1 ppm + 2, 4-D 0.1 ppm, the highest induction frequency of callus is 90.4% and from which the buds differentiation frequcncy is 23.8%, the response of different parts of scape is si- milar. Histocytological observations on explants indicated that the callus was induced from cells of cortex, phloem and peripheral zone of vascular bundle. After inocubation of explants for one month, adventitious huds were formed from either the cells of surface layer of callus or leaf-likc cylindrica1 structure derived from callus.
